Make sure to ALWAYS, ABSOLUTELY ALWAYS, WITHOUT FAIL, use the highest quality serums.
QFT, but also note that although "excellent" quality is the best you'll see, having a serum reach that point doesn't mean it's the "best" - make sure the purifier says it can't be made better before you try to put it to use.

Using bee serums is actually a lot easier than manually breeding duplicate bees once you get the hang of it.
This depends on what you want to duplicate. If you want to turn a princess from a freshly-scooped hive into a clone of a max speed/territory/tolerance/etc princess, you can either spend forever injecting serums into it, or you can just let it breed a few times with a stack of suitable drones to bring multiple traits into line at once.

If you want to bypass the annoying hybrid stage involved in mutating to each new species, serums are your best friend for skipping through that process.

I wrote out this post explaining the genetic machines, once upon a time. Might be useful. Reading this thread will also answer a lot of questions related to bee-keeping.

Mostly it's all been said already... I would just add that once you have max fertility serum it helps to use it to access more drones for isolating, I always add that after I get my first princess / drone mutation - that way I am guaranteed 4 drones, 1 drone & the princess go back into an an apiary / alviary & the other 3 drones go into the isolator - you have a very good chance of getting the species serum from those 3 drones. Then I fall back on the 100's of Rocky princess' & drones as a base for injecting new species serums into & making a solid production pair (add whatever serum traits over & above), there's far less additional inoculating required as the Rocky's already have nocturnal, rainfall, cave dwelling & both 2 humidity & temperature. Don't forget to inoculate the princess & drones before they breed to a queen, can get a little messy otherwise & cost you a few hours of time. I'm not sure if my method is close to efficient, but its pretty dependable.

what I often see people doing is putting their bee species constantly in the acclimatiser to change their temp/hum tolerance, but you can actually just put 1 bee in the acclimatiser, get it to the highest temp/hum tolerance, and make serums from that.

You don't even need an acclimatizer for that. Rocky bees naturally have +2 both temperature and humidity tolerances, just extract the trait from them.

That's not enough to use all bees in every biome though. And +5 both is easy to reach in an acclimatizer.

If you only acclimatise one bee, then odds are the isolator will kill it before you get the serums you want. However, the acclimatiser can upgrade the tolerances on an entire stack of drones at once, so put a bunch of them in there in one go and max them all out before going off to get your serums.

Once you have those serums, it's indeed time to retire your acclimatiser.
