    You don't have to be in a specific biome, but you do have to have a clear path to the sky from the water block. I'm guessing thats what was causing all the trouble. It's supposed to simulate a small pond with wind to carry seeds and sunlight to sprout them.

    According to your crash log, you are using Ex Nihilo version 1.28. Agrarian Skies is currently using Ex Nihilo version 1.29d. I fixed this a long time ago. You need to update to a newer version of the mod pack.

    You'll need to trigger a whole bunch of simultaneous spread actions. Maybe fly over a jungle biome infesting as you go and wait about 20 seconds?
