Is 3.75 Gigs of RAM enough to fully run the Direwolf20 1.6 pack?

If not, what pack should I run without having to worry about the capacity of my RAM?

That's more than enough, you should be fine. 2.5-4gb is what I consider the optimal amount of RAM ;)

Are we talking total RAM or allocated?
You will want to allocate around 2GB in the launcher for the game. 2.5GB if you are running a texture pack. This will not leave an awful lot for your OS and the fact that the java client usually spends double of what you allocate. But should be playable, specially if you play on a server and not singleplayer.
 
Oh, I am talking about allocated.

I play usually in Singleplayer due to the fact I find servers too laggy to play on.

Ok then don't allocate more than 2.5ish for DW20. You don't gain anything from allocating more, quite the contrary you only allow java to fuck up more. Generally allocate the least amount of RAM possible while still being able to play the game. 2GB without texture pack, 2.5GB with texture pack should fit the size of DW20 just fine.
